乌头汤对膝骨关节炎TLR4信号通路的影响

摘要

目的观察乌头汤对膝骨关节炎(Kneeosteoarthritis,KOA)大鼠滑膜组织TLR4信号通路关键因子(TLR4/MyD88/NF-kB)的调控作用,从而探讨其作用机制。方法取36只大鼠随机分为空白组(不进行任何处理)、模型组(膝关节骨性关节炎造模后生理盐水灌胃)、治疗组(膝关节骨性关节炎造模后乌头汤灌胃),干预2个月后比较各组大鼠膝关节滑膜组织特征指标差异,并观察大鼠膝关节关节软骨病理形态变化。结果:(1)大鼠滑膜组织中诱导型一氧化氮合酶(iNOS)、肿瘤坏死因子α(TNFα)、白细胞介素6(IL-6)的表达与空白组相比,模型组iNOS、TNFα和IL-6质量浓度显著升高(均P<0.05);治疗组iNOS、TNFα和IL-6质量浓度均显著升高(均P<0.05)。与模型组相比,治疗组iNOS、TNFα和IL-6质量浓度均显著降低(均P<0.05)。(2)相比空白组,模型组TLR4、TRAF6、NF-kBp65均有显著升高(均P<0.05);相比模型组,治疗组TLR4、MyD88、TRAF6、NF-kBp65均有下降(均P<0.05)。结论乌头汤通过影响TLR4信号通路改善膝关节功能障碍程度,提高关节活动度。
